| ProductName: | | PKC theta Antibody |
| Product Description: | | Rabbit Polyclonal anti-PKC theta [Ser695] |
| Clonality: | | Polyclonal |
| Immunogen: | | Synthetic phosphopeptide derived from a region of human PKC theta that contains serine 695. |
| Specificity: | | This antibody does not cross-react with any other PKC isoforms tested. |
| CrossReactivity: | | Cross-reacts with Human. Expected to cross-react with Mouse (75% identity with immunogen) due to sequence homology. Not yet tested in other species. |
| Packaging: | | 0.1 ml Immunogen affinity purified Rabbit antisera. |
| Uses: | | WB: Use at a concentration of 0.1 - 1.0 ug/ml. Detects a band of approximately 80 kDa. Not tested in other applications. Optimal dilutions/concentrations should be determined by the end user. |
| Control: | | Jurkat cells treated with PMA, a phorbol ester. |
| Background: | | Protein Kinase C theta (PKC theta) is an 80 kDa member of the novel group (nPKCs: sensitive to diacylglycerol, phosphatidylserine and phorbol esters) of the PKC family of serine/threonine kinases that are involved in a wide range of physiological processes including mitogenesis, cell survival and homeostasis. Transgenic mice over-expressing dominant negative PKC theta develop hyperinsulinimia. PKC theta is involved in JNK activation and also plays a specialized role in TCR mediated activation of T and B cells. The activation of PKC theta in T cells is associated with its recruitment to the membrane, and is mediated by PI3-kinase and Vav. PKC theta is a constitutively competent kinase and is auto-phosphorylated on serine 676 in the turn loop and serine 695 in the hydrophobic loop. The phosphorylation on serine 695 contributes to the catalytic activity of PKC theta. |
