| Specificity: | This antibody is specific for human IRS 2 phosphorylated at the position of Serine 731. The antibodies were evaluated for specificity with a dot blot assay using synthetically prepared IRS peptides. It only recognizes the phosphorylated serine 731, not other phosphorylated sites or non-phosphorylated IRS 1 or IRS 2. |

| Uses: | Immunoprecipitation: use at a concentration of 3.0 - 5.0 ug / extract from 10<sup>7</sup> cells. Western Blotting: use at a concentration of 0.5 - 2.0 ug/ml. ELISA: use at a concentration of 0.1 - 1.0 ug/ml. Not tested in other applications.Optimal dilutions / concentrations should be determined by the end user. By Western blot, this antibody detects a band of approximately 200 kDa in cell lysate derived from mouse serum-treated fibroblasts, which corresponds to the predicted molecular weight of phospho IRS 2. Samples were boiled prior to subject 7.5% SDS-polyacrylamide gel electrophoresis. |
| Background: | The insulin receptor mediates insulin action through the phosphorylation of substrate proteins. The major substrates of the insulin receptor kinase are the insulin receptor substrate (IRS) family of proteins, IRS 1, IRS 2, IRS 3, IRS 4. IRS 2, a cytoplasmic signaling molecule that mediates effects of insulin, Insulin-like growthfactor 1 and other cytokines by acting as a molecular adaptor between diverse receptor kinases and downstream effectors. IRS proteins contain more than 70 potential Ser / Thr phosphorylation sites for kinases like PKA (cAMP dependent protein kinase), PKC, and MAPK. The phosphorylation of Ser / Thr residues of IRS proteins has adual function, serving either for a positive or negative modulation of insulin signal transduction. |
