| Background: | STATs, such as STAT5, are proteins that serve the dual function of signal transducers and activators of transcription in cells exposed to signaling polypeptides. In response to cytokines and growth factors, STAT family members are phosphorylated by the receptor associated kinases, and then form homo- or heterodimers that translocate to the cell nucleus where they act as transcription activators. STAT5 is activated by, and mediates the responses of many cell ligands, such as IL2, IL3, IL7 GM-CSF, erythropoietin, thrombopoietin, and different growth hormones. STAT5a and STAT5b knockout mice show important roles in prolactin-mediated mammary gland development and growth hormone-mediated induction of sexual dimorphism, respectively. Abnormal activity of certain STAT family members, particularly STAT3 and STAT5, is associated with a wide variety of human malignancies, including hematologic, breast, head and neck, and prostate cancers. STAT5A and STAT6, are selectively activated when the heart is subjected to ischemic injury, whereas activation of STAT3 and STAT5A is involved in myocardial hypertrophy. STAT5 and STAT3, as these two STAT molecules are required for normal breast development and involution, respectively, and may play an important role in breast carcinogenesis. |
