| Uses: | Peptide ELISA: antibody detection limit dilution 1:32,000. Western Blot: Preliminary experiments gave an approx 150-170kDa band in Human Liver and Pancreas lysates after 1µg/ml antibody staining. Please note that currently we cannot find an explanation in the literature for the band we observe given the calculated size of 99.9kDa according to NP_006199.1. The 150kDa band was successfully blocked by incubation with the immunizing peptide. We would appreciate any feedback from people in the field - have any results been reported with other antibodies/lysates? Have any further splice variants/modified forms been reported?Immunohistochemistry: In paraffin embedded Liver shows membranous staining in many hepatocytes. Recommended concentration, 1-2µg/ml. |
| Background: | ENPP1 has a broad specificity and cleaves a variety of substrates, including phosphodiester bonds of nucleotides and nucleotide sugars and pyrophosphate bonds of nucleotides and nucleotide sugars. It can hydrolyze nucleoside 5' triphosphates such as ATP, GTP, CTP, TTP and UTP to their corresponding monophosphates with release of pyrophosphate. It can also hydrolyze diadenosine polyphosphates and 3',5'-cAMP to AMP. It may play a role in the regulation of pyrophosphate production, the regulation of the availability of nucleotide sugars in the endoplasmic reticulum and Golgi, and the regulation of purinergic signaling. |
