Uses:Immunoprecipitation (denatured, with protein G): use at a concentration of 2ug / mg of lysate.Western Blotting: use at a concentration of 2 - 4ug / ml for 2hrs at RT.Immunohistochemistry (formalin fixed paraffin embedded): use at a concentration of 1 - 2ug / ml for 30 min at RT.Optimal dilutions / concentrations should be determined by the end user.In Western blot, this antibody detects a band of approximately 55 kDa, which corresponds to Caspase 8.For formalin fixed paraffin embedded sections, perform heat mediated antigen retrieval using 10mM citrate buffer pH 6.0 for 10 - 20 minutes before commencing with the IHC staining protocol.

Background:Caspases are cysteine proteases, expressed as inactive precursors, that mediate apoptosis by proteolysis of specific substrates. Caspases have the ability to cleave after aspartic acid residues. There are two classes of caspases involved in apoptosis; initiators (activation by receptor cluster) and effectors (activation by mitochondrial permeability transition). Proapoptotic signals autocatalytically activate initiator caspases, such as Caspase 8 and Caspase 9. Activated initiator caspases then process effector caspases, such as Caspase 3 and Caspase 7, which in turn cause cell collapse.
