| Uses: | Immunofluorescence: assay dependent.Immunoprecipitation (with protein G):use at a concentration of 2ug/mg of protein lysate (native and denatured).Western Blotting: use at a concentration of 1 - 2mg/ml for 2hrs at RT.Immunohistochemistry (formalin fixed paraffin embedded / frozen): use at a concentration of 2 - 4mg/ml for 30 min at RT.Not tested in other applications.Optimal dilutions / concentrations should be determined by the end user.By Western blot, this antibody detects a band of ~ 460 kDa, which corresponds to the predicted molecular weight of DNA-PKcs. This antibody reacts with intact DNA-PKcs as well as its degradation products. DNA-PKcs inhibits ~50% of the kinase activity of DNAPKcs. Preincubation of enzyme with DNA protects DNA-PKcs against inactivation by this antibody.Staining of formalin-fixed tissues requires boiling tissue sections in 10mM citrate buffer, pH 6.0,for 10-20 min followed by cooling at RT for 20 min. |

| Background: | DNA PK is the key component of the non-homologous end-joining (NHEJ) pathway of DSB repair in mammalian cells. DNA PK consists of a heterodimeric DNA-binding subunit (Ku70/80) and an approximately 465 kDa catalytic subunit (DNA PKcs). DNA PKcs is a serine/threonine protein kinase whose activity is greatly stimulated by its recruitment to DNA breaks by the Ku heterodimer. |
