| Uses: | This antibody can be used for ELISA and Western Blot. In Western Blot, Preliminary experiments gave an approx 25kDa band in HeLa lysates after 1ug/ml antibody staining. Please note that currently we cannot find an explanation in the literature for the band we observe given the predicted size of 75kDa according to NP_031374. The 25 kDa band was successfully blocked by incubation with the immunizing peptide. Not tested in other applications. ATF6, a basic-leucine zipper protein, was isolated by yeast interaction assay as a binding partner to SRF (serum response factor) (1). SRF is a transcription factor, which binds to the serum response element (SRE) in the c-fos promoter and regulates expression of the c-fos gene as well as other immediate-early genes and some tissue-specific genes. ATF6 has also been shown to interact with the endoplasmic reticulum (ER) stress response element (ERSE) (2). ATF6 constitutively expressed as a membrane-bound 90-kDa protein is directly converted to a 50-kDa protein in ER-stressed cells (3). When unfolded proteins accumulate in the ER, ATF6 is released and translocates to the golgi where it is cleaved into its active form which enters the nucleus and activates translation of UPR genes including XBP1 (4).] |
