Uses:This antibody can be used for ELISA and Western Blot. In Western Blot preliminary experiments gave approx 50kDa and 60kDa bands in Human Muscle lysates. These bands were successfully blocked by incubation with the immunising peptide. Please note that currently we cannot find an explanation in the literature for the bands we observe given the predicted size of approx. 110kDda according to NP_004454. Not tested in other applications.Recommended Starting Dilution:*ELISA (peptide): 1:1,000WB: 1ug/ml *Optimal dilutions should be determined by the end user.
Background:**Please note, this product is one of a range of Investigative Grade antibodies, made against targets that have limited or no commercial antibodies available to them and for which there are no data on the expression of the protein in the range of common cell lines and tissues available to us. These antibodies are affinity purified using their peptide immunogen and are known to give low background staining in a western blot. However no additional claims are made for their ability to recognize native protein in any application. FGD1 contains Dbl (DH) and pleckstrin (PH) homology domains. It can bind specifically to the Rho family GTPase Cdc42Hs and stimulate the GDP-GTP exchange of the isoprenylated form of Cdc42Hs. It also stimulates the mitogen activated protein kinase cascade leading to c-Jun kinase SAPK/JNK1 activation. FGD1 has an essential role in embryonic development, and FGD1 gene mutations result in the human developmental disorder, Aarskog-Scott syndrome.
