Specificity:CD44 is a transmembrane glycoprotein with Mr=80 kDa. CD44 is found primarily on granulocytes, mature T lymphocytes and macrophages as well as most other cells of leucocyte origin except cortical thymocytes. CD44 is also expressed by red blood cells, fibroblasts, epithelial cells, and cells of the brain. CD44 serves as the receptor for hyaluronan, the major glycosaminoglycan of the extracellular matrix. CD44 mediates epithelial cell attachment to basement membranes. CD44 also mediates lymphocyte attachment to and rolling on endothelial cells, lymphocyte homing to lymphoid organs and inflammatory sites, and lymphocyte aggregation. Recent evidence indicates that stimulation of cells by hyaluronan causes the protein kinase Src to be recruited to CD44-containing complexes, an event which results in increased phosphorylation of Src tyrosine 418, increased Src kinase activity, increased phosphorylation of the Src substrate cortactin, and changes in the organization of the cytoskeleton. This antibody binds to epitope 1 of CD44.

Uses:FACS: Use 0.5-1ug for 10<sup>6</sup> cells.IHC-P: Use at a concentration of 0.5 - 2 ug/ml. Antigen retrieval is not essential but may optimise staining.IHC-Fr: Use at a concentration of 0.5 - 2 ug/ml. IP: Use at a concentration of 0.5 - 2 ug/ml. Not tested in other applications.Optimal dilutions/concentrations should be determined by the end user.

Background:CD44 is a type 1 transmembrane glycoprotein also known as Phagocytic Glycoprotein 1 (pgp 1) and HCAM. CD44 is the receptor for hyaluronate and exists as a large number of different isoforms due to alternative RNA splicing. The major isoform expressed on lymphocytes, myeloid cells, and erythrocytes is a glycosylated type 1 transmembrane protein. Other isoforms contain glycosaminoglycans and are expressed on hematopoietic and non hematopoietic cells. CD44 is involved in adhesion of leukocytes to endothelial cells, stromal cells, and the extracellular matrix.
Storage:Store at 2-8°C for up to one month. For long term storage, apportion into working aliquots and store at -20°C. Avoid repeated freeze-thaw cycles.
