| Background: | The activation of the initiator caspase, caspase 9, is regulated by the apoptosome, which is composed of cytochrome c bound to Apaf1 and dATP. The apoptosome responds to a diverse array of apoptotic stimuli that initiate the caspase cascade that ultimately results in cell death. The cascade of events starts with the proteolytic cleavage of caspase 9, that then cleaves downstream caspases, caspase 3, 6, and 7. The apoptotic initiating activity of caspase 9 can be regulated by XIAP, X-linked inhibitor-of-apoptosis protein; through the binding of caspase 9 at its amino terminus while it is bound to the apoptosome. This inhibition of caspase 9 activity has been shown to be relieved by Smac/DIABLO through its interaction with XIAP. |
