| Background: | | Proline-rich/Ca2+ -activated tyrosine kinase (PYK 2, also known as RAFTK, CAK beta or Focal Adhesion Kinase 2) is a member of the Focal Adhesion Kinase family of non-receptor, proline-rich PTK's. PYK 2 signalling is initiated by a variety of extracellular stimuli including integrin ligation, CD surface marker ligation (e.g., CD3, CD28, TCR, VCAM), bioactive peptides, growth factors, cytokines, chemokines, and certain stress stimuli. PYK 2 is involved in the regulation of vesicular transport, osteoclastic bone resorption, modulation of ion channels, T- and B-cell receptor signaling and cell death. Phosphorylation of the catalytic domain tyrosines 579 and 580 results in maximum activation of PYK 2. Tyr579 and Tyr580 are phosphorylated by Src. |
