| Background: | Dok 2/Dok R (downstream of tyrosine kinases, aka p56dok) is a 56 kDa protein and a member of the Dok family of RasGAP binding proteins. Dok-2 plays an important role downstream of several growth factor receptors, including VEGFR and EGFR, and is implicated in negative regulation of signaling pathways activated by tyrosine kinases. Dok-2 associates with Tek following angiopoietin-1 stimulation, resulting in tyrosine phosphorylation of Dok 2 and the subsequent recruitment of Nck and the p21 activating kinase (PAK/PAK1) to the activated receptor. Dok 2 can also be tyrosine phosphorylated by Lyn, ErbB-2/Neu, Bcr/Abl and rasGAP. Dok 2 tyrosine 142 is conserved with Dok 1 tyrosine 146, a phosphorylation site that inhibits Src transformation. |
