| Background: | Filaggrins are an important class of the intermediate filament-associated proteins which interect with keratin intermediate filaments (Ifs) of terminally differentiating mammalian epidermis. Filaggrins exhibit wide species variations. Filaggrin is expressed only in well differentiated keratinized epithelial cells. The synthesis of filaggrin varies in types of ichthyosis, and with viral, premalignant and malignant conditions. Filaggrin also appears to be a target molecule for rheumatoid arthritis-specific auto-antibodies in humans. Keratohyalin granules in the granular layer of interfollicular epidermis are predominantly composed of the 400-kD protein profilaggrin. Following a short, unique N-terminal domain, most of the profilaggrin molecule consists of 10 to 12 repeats of the 324-residue filaggrin sequence. Upon terminal differentiation of granular cells, profilaggrin is proteolytically cleaved into filaggrin peptides of approximately 37 kD and the N-terminal domain containing an S100-like calcium-binding domain. |
