| Background: | Thrombospondin (TSP) is a major glycoprotein constituent of the á-secretory granules of human platelets. In its native state it has a molecular weight of 420 kDa and consists of 3 identical disulfide-linked subunits. Thrombospondin can bind to heparin, fibrinogen, collagen, fibronectin, plasminogen and histidine-rich glycoproteins. Together with fibrinogen TSP may mediate aggregation of platelets activated by low concentrations of thrombin. In addition to megakaryocytes, TSP can also be synthesized by vascular endothelial cells, fibroblasts, smooth muscle cells, monocytes, granular pneumocytes and astrocytic neuroglial cells in tissue culture. The physiological role f TSP is still poorly understood although it has been postulated to be involved in platelet aggregation and cellular adhesion. Because TSP is secreted only after platelets are activated and the fact that it has a much longer intravascular half life than other platelet specific proteins, there is an increasing interest in using the plasma level of TSP as a clinical indicator of intravascular platelet activation. |
