| Uses: | | Dot: concentration determined using recombinant NGFB at 400 ng under non- and 200 ng under reducing conditions. For capture ELISA, Monoclonal Anti-Human Nerve Growth Factor-Beta can be used as the capture antibody. Determined in combination with a biotinylated NGF-Beta polyclonal detection antibody at 50-100 ng/ml (100 ul/well), an ELISA for sample volumes of 100 ul with a range of 31.2 to 2000 pg/ml may be obtained. For capture ELISA, a working concentration of 1-2 ug/ml is determined to coat plates at 100 ul/well.Inhib: Use at a concentration of 1 - 2 ug/ml. Human NGFB stimulates the 3H-thymidine incorporation by TF-1 cells in a dose-dependent manner. The Neutralization Dose50 (ND50) for anti-human NGFB is approximately 1-3 ug/ml in the presence of 5 ng/ml of recombinant human NGFB using the NGFB responsive cell line TF-1. The ND50 is the concentration of antibody required to yield one-half maximal inhibition of the cytokine activity on a responsive cell line, when the cytokine is present at a concentration just high enough to elicit a maximum response. The exact concentration of antibody required to neutralize human NGFB activity is dependent on the cytokine concentration, cell type, growth conditions, and the type of activity studied. |
