Immunogen:Synthetic peptide: SDSPREQDSESQTLDK conjugated to KLH, corresponding to N terminal amino acids 209-224 of Human Caspase 8.
Epitope:The immunogen sequence corresponds to the N-terminal region of the enzyme p20 subunit and is identical in human caspase 8 isoforms (caspase8b/MACH alpha 2, MACH beta 3 and MACH beta 4).
Specificity:Anti-Caspase 8 (SK-16) recognizes human caspase 8a and caspase 8b by immunoblotting. The product may also detect the caspase 8 MACH beta 3/beta 4 isoforms.
Cross Reactivity:Cross-reacts with Human.Expected to cross-react with Mouse (85% identity with immunogen) due to sequence homology.Not yet tested in other species.

Uses:WB: Use at a dilution of 1/1000 (determined by using a whole cell extract of the human Burkitt lymphoma Raji cell line). Expected bands at: 55 kDa (human caspase 8a) and 54 kDa (caspase 8b). Possible bands at 30-32 kDa (caspase 8 MACH beta 3/beta 4 isoforms). Not tested in other applications.Optimal dilutions/concentrations should be determined by the end user.

Background:Caspases are cysteine proteases, expressed as inactive precursors, that mediate apoptosis by proteolysis of specific substrates. Caspases have the ability to cleave after aspartic acid residues. There are two classes of caspases involved in apoptosis; initiators (activation by receptor cluster) and effectors (activation by mitochondrial permeability transition). Proapoptotic signals autocatalytically activate initiator caspases, such as Caspase 8 and Caspase 9. Activated initiator caspases then process effector caspases, such as Caspase 3 and Caspase 7, which in turn cause cell collapse.
