Immunogen:Synthetic peptide: KPDRSSFVPSLFSKKKKN-C conjugated to maleimide-activated BSA, corresponding to amino acids 25-42 of Human Caspase 7 with C-terminal added cysteine.
Epitope:Located at the N-terminal region of caspase 7 (amino acid residues 25-42 with C-terminal added cysteine). The epitope also corresponds to amino acid residues 2-19 of the human enzyme p20 subunit.
Specificity:The antibody reacts specifically with procaspase 7 (Mch3).
Cross Reactivity:Cross-reacts with Human.Expected to cross-react with Mouse (82% identity with immunogen) and Rat (85% identity with immunogen) due to sequence homology.Not yet tested in other species.

Uses:WB: Use at a dilution of 1/1000 (determined by using a whole extract of a Jurkat human T-cell leukemia cell line). Predicted molecular weight: 35 kDa. An additional weak band, representing a caspase 7 degradation product, may be detected in some cell line extracts.Not tested in other applications.Optimal dilutions/concentrations should be determined by the end user.

Background:Caspases are cysteine proteases, expressed as inactive precursors, that mediate apoptosis by proteolysis of specific substrates. Caspases have the ability to cleave after aspartic acid residues. There are two classes of caspases involved in apoptosis; initiators (activation by receptor cluster) and effectors (activation by mitochondrial permeability transition). Proapoptotic signals autocatalytically activate initiator caspases, such as Caspase 8 and Caspase 9. Activated initiator caspases then process effector caspases, such as Caspase 3 and Caspase 7, which in turn cause cell collapse.
Storage:Store at 4°C short term (1-2 weeks). Aliquot and store at -20°C or -80°C. Avoid repeated freeze / thaw cycles.
