| Background: | The secreted polypeptide noggin (encoded by the Nog gene) binds and inactivates members of the transforming growth factor beta superfamily of signaling proteins (TGFbeta-FMs), such as BMP4. By diffusing through extracellular matrices more efficiently than TGFbeta-FMs, noggin may have a principal role in creating morphogenic gradients. Findings report that noggin is essential for joint formation and suggest that NOG requirements during skeletogenesis differ between species and between specific skeletal elements within species (1). Noggin is a bone morphogenetic protein (BMP) antagonist expressed in Spemann's organizer. Murine Noggin is expressed in condensing cartilage and immature chondrocytes, as are many BMPs (2). The structure of the antagonist Noggin bound to BMP-7, shows that noggin inhibits BMP signaling by blocking the molecular interfaces of the binding epitopes for both type I and type II receptors (3). |
