| Background: | Parkin is a RING-type E3 ubiquitin-protein ligase which binds to E2 ubiquitin-conjugating enzymes. Parkin is composed of a ubiquitin-like domain at its amino terminus and two RING finger motifs flanking a cysteine-rich region that is known as the in between RING fingers (IBR) motif (1-2). Involved in the ubiquitination pathway for misfolded proteins derived from endoplasmic reticulum, Parkin contributes to protection from neurotoxicity induced by unfolded protein stresses. Parkin can also ubiquitinates itself to promotes its own degradation (3).Defective Parkin is involved in Parkinson's diseases and in autosomal recessive juvenile Parkinsonism (AR-JP) (4-5) |
