| Background: | IRAK-3 (also known as IRAK-M) is a member of the interleukine-1 receptor-associated kinase (IRAK) family. The IRAK family is implicated in the Toll-like receptor (TLR) and Il-1R signaling pathway (1). Upon stimulation, IRAK gets phosphorylated and activates NF-kB (2). Unlike IRAK, IRAK2 and IRAK4 which are expressed ubiquitously, IRAK-3 is mostly expressed in monocytes and macrophages (3). IRAK-M and IRAK-2 lack kinase activity, while IRAK-3 has a weak capacity to be phosphorylated. Additionally, IRAK-3 has been found to negatively regulate TRL signaling to protect from endotoxic shock (5). |
