| Background: | Apaf-1, or apoptotic protease activating factor 1, is a human protein homolog of the C. elagans apoptotic regulatory protein CED-4 that activates caspase-9 during apoptosis (1). Cytosolic Apaf-1 forms an oligomeric complex with caspase-9 in the presence of cytochrome c and dATP, which leads to caspase-9 activation and subsequent activation of caspase-3 (2, 3). Oligomeric Apaf-1 and pro-caspase-9 bind to each other via their respective NH2-terminal CARD domains, consequently enhancing the catalytic activity of caspase-9 (4), leading to activation of the caspase signaling cascade and apoptosis. |
