Uses:AP: Use at an assay dependent concentration. ELISA: Use at an assay dependent dilution.IA: Use at an assay dependent concentration. WB: Use at an assay dependent concentration. These antibody combinations are recommended for sandwich ELISA, and demonstrate high sensitivity (0.02 ng/ml), good kinetics, low background and high reproducibility: detection. Using multiple combinations of MAbs antibodies with different epitope specificity as capture and multiple MAbs as detection, the sensitivity of the sandwich ELISA can be increased 2-4 fold. The ELISA based on these recommended Abs, recognize complexed and free forms of the antigen with same sensitivity. For better sensitivity and reproducibility for assay designing we recommend to use at least one antibody that recognizes the stable fragment of the cTnI molecule (30-110 aa section), as cTnI is proteolytically degraded in the blood stream.Because the amount of complexed cTnI in blood of AMI patients is very high, for cTnI immunodetection we recommend using also a MAb that is specific to TnC or a MAb specific to TnC together with MAb(s) specific to cTnI. Such approach helps improving the sensitivity and reproducibility of cTnI immunoassays.
Background:Equally reactive with free cardiac troponin I (cTnI) and cTnI forming complexes with other troponin components. (In the presence of 5 mM EDTA). Antibodies are not affected by heparin, phosphorylation, oxidation and troponin complex formation. No cross-reaction with skeletal muscle troponin I.
