| Background: | CaMKII (also known as calcium/calmodulin-dependent kinase II) is a serine/threonine kinase and a member of the Ca2+/calmodulin-dependent protein kinase subfamily. This enzyme is composed of two major neuron-specific subunits (alpha and beta) and two subunits expressed in non-neuronal tissues (delta and gamma). The alpha subunit is approximately 50 kD. CAMKII is prominent in the central nervous system and widely expressed in both the adult and fetal brain. The CAMKII kinase phosphorylates substrates in the brain including synapsin I and nitric oxide synthase in response to increases in intracellular free calcium; autophosphorylation at Thr286 permits CaMII kinase to be active in the absence of free calcium. The alpha subunit interacts with the beta subunit of CaMII kinase and associates with the NR2B subunit of the NMDA receptor. CAMII kinase is believed to be involved in the consolidation of long-term memories in the brain and synaptic plasticity. The Poly6182 antibody has been shown to be useful for Western blotting of human phosphorylated CAMII kinase (Thr286). |
