| Background: | PTEN (also known as phosphatase and tensin homolog deleted on chromosome ten and MMAC1) is a protein-tyrosine phosphatase non-receptor containing a tensin domain and PDZ binding domain. This 47 kD protein is found in the cytoplasm and functions as a potential tumor suppressor. PTEN is a negative regulator of the PI3K/AKT signaling pathway regulating phosphoinositide(3,4,5)P3 levels, and an inhibitor of cell migration. PTEN is mutated in numerous cancers. PTEN activity is inhibited by phosphorylation and is transcriptionally upregulated by PPAR gamma agonists. PTEN has been shown to interact with phosphatidylinositol 3,4-bisphosphate, PtdIns(3,4,5)P(3), MAGI-1, MAGI-2, MAGI -3, hDLG, and MAST205. The Poly6144 antibody recognizes the N-terminal region of human PTEN and has been shown to be useful for Western blotting and immunofluorescence staining. |
