| Background: | NCOA1/SRC1 (nuclear receptor coactivator 1/steroid receptor coactivator 1) is a 157 kD member of the steroid receptor coactivator family containing a nuclear localization sequence, bHLH, and PAS domain. Three isoforms of this nuclear protein have been reported. NCOA/SRC1 is expressed in astrocytoma, melanoma cell lines, lymphocytes, mammary gland, and skeletal muscle. This protein exhibits histone acetyltransferase activity, and is a transcriptional coactivator for steroid and nuclear hormone receptors. NCOA/SRC1 is inhibited by constitutively activated androstane receptor (CAR) signaling and activated by thyroid hormone receptor, and estrogen receptor ligation. NCOA/SRC1 is modified by phosphorylation and sumoylation; phosphorylation enhances coactivator function.This protein forms a complex with NCOA3, IKKA, IKKB, IKBKG and histone acetyltransferase protein CREB binding protein, and has been shown to interacts with HIF1A, APEX, NR3C1, NR3C2, STAT6, STAT3, and the androgen receptor. |
