| Uses: | Each lot of this antibody is quality control tested by immunofluorescent staining with flow cytometric analysis. For immunofluorescent staining, the suggested use of this reagent is: 1.0 microgram per one million cells in 100 microliter volume. It is recommended that the reagent be titrated for optimal performance for each application. |
| Background: | CD222 is a ubiquitously expressed 250 kD transmembrane protein found on the cell surface and in the late endosomal/prelysosomal compartment. It is a receptor for insulin-like growth factor II (IGF II) and mannose-6-phosphate. Binding of IGF II initiates a broad range of biologic responses including cell growth, adhesion, and angiogenesis. CD222 has also been shown to internalize lysosomal enzymes and to bind and to activate latent TGF beta. In addition, CD222 interacts with a variety of extracellular and intracellular ligands and associates with other membrane proteins including granzyme B, plasminogen activator urokinase, aspartylglucosaminidase, phosphofurin acidic cluster sorting protein 1, cathepsin B, D, and L, retinoic acid, CD87, and TGF beta receptor-V. CD222 can be modified by phosphorylation, glycosylation, and palmitoylation and has been shown to undergo proteolytic cleavage from the cell surface to produce a soluble CD222 protein. Mutations in CD222 have been implicated in mucolipidosis and are also observed in hepatocellular carcinoma. The MEM-238 antibody is useful for flow cytometry and Western blotting. |
