| Specificity: | NB100-66277 recognises p21-activated kinase PAK6, a 75kDa, serine/threonine protease expressed in the testis and prostate. p21-activated kinases (PAKs) bind activated Cdc42 and Rac via a Cdc42/Rac interactive binding (CRIB) domain. Group 1 PAK kinases (PAK1, 2 and 3) regulate the activity of an adjacent conserved autoinhibitory domain and are associated with cellular processes, including cytoskeleton rearrangement, apoptosis and MAP kinase signalling. Group 2 PAKs (PAK4, 5, and 6) lack the autoinhibitory domain and are not activated by Cdc42/Rac binding.PAK6, in the presence of dihydrotestosterone, interacts with androgen receptor (AR), a steroid hormone-dependent transcription factor involved in male sexual differentiation and development. On androgen stimulation, PAK6 cotranslocates into the nucleus with AR. PAK6 is activated by MAP kinase implying that it is involved in the response to stress signals. |

| Background: | The p21 activated kinases (PAK) are critical effectors that link Rho GTPases to cytoskeleton reorganization and nuclear signaling. The PAK proteins are a family of serine/threonine kinases that serve as targets for the small GTP binding proteins, CDC42 and RAC1, and have been implicated in a wide range of biological activities. The protein encoded by this gene is activated by proteolytic cleavage during caspase-mediated apoptosis, and may play a role in regulating the apoptotic events in the dying cell. |
