| Specificity: | NB100-66276 recognises p21-activated kinase PAK4, a 64kDa, serine/threonine protease expressed in the testis, prostate and colon. p21-activated kinases (PAKs) bind activated Cdc42 and Rac via a Cdc42/Rac interactive binding (CRIB) domain. Group 1 PAK kinases (PAK1, 2 and 3) regulate the activity of an adjacent conserved autoinhibitory domain and are associated with cellular processes, including cytoskeleton rearrangement, apoptosis and MAP kinase signalling. Group 2 PAKs (PAK4, 5, and 6) lack the autoinhibitory domain and are not activated by Cdc42/Rac binding.PAK4 binds activated Cdc42, weakly activating the Jun N-terminal kinase signalling pathway. Binding of Cdc42 by PAK4 also results in their colocalisation to the Golgi where PAK4 mediates the formation of actin-enriched filopodia protrusions. PAK4 may also stabilise actin filaments by inactivating protein phosphatase SSH1, a phosphatase which releases the actin binding/depolymerising factor cofilin from inhibition allowing it to destabilise actin filaments. |

| Background: | The p21 activated kinases (PAK) are critical effectors that link Rho GTPases to cytoskeleton reorganization and nuclear signaling. The PAK proteins are a family of serine/threonine kinases that serve as targets for the small GTP binding proteins, CDC42 and RAC1, and have been implicated in a wide range of biological activities. The protein encoded by this gene is activated by proteolytic cleavage during caspase-mediated apoptosis, and may play a role in regulating the apoptotic events in the dying cell. |
