| Specificity: | NB100-63220 recognises human phosphatase and tensin homolog (mutated in multiple advanced cancers 1) (PTEN), a 47 kDa tumour suppressor expressed in all adult tissues. PTEN inhibits the AKT/PKB signalling pathway by dephosphorylating phosphoinositides which then associate with programmed cell death 6 interacting protein, suppressing AKT1. PTEN dephosphorylates tyrosine-phosphorylated focal adhesion kinase, inhibiting cell migration, integrin-mediated cell spreading and focal adhesion formation. It may also negatively regulate insulin signalling and glucose metabolism in adipose tissue. Defects in PTEN are associated with Cowden disease, Lhermitte-Duclos disease, Bannayan-Zonana syndrome and susceptibility to various cancers. |
