| Specificity: | NB100-63218 is specific for PRK-1. It does not recognise other human kinases such as AKT1, AKT2, TSF1, TSK1, EphB4 or CSK in Western blotting or ELISA.PRK1 is a ubiquitously expressed monomeric protein consisting of an N-terminal regulatory domain and a C-terminal catalytic domain. The activation loop at the catalytic domain of inert PRK1 is phosphorylated by the upstream kinase, PDK1, causing it to assume a catalytically competent but inactive confirmation. The binding of effector molecules, such as GTP-RhoA and various phosphol-ipids, causes a conformational change resulting in activation of PRK1 by exposing the active site. It is speculated that the activation of PRK1 by PDK1 may mediate insulin signals to the actin cy-toskeleton. The proteolytic activation of PRK1 by caspase-3 or related proteases during apoptosis suggests its role in signal transduction related to apoptosis. Alternatively spliced transcript variants encoding distinct isoforms have been observed. |
