Background:Principal Names: Wingless-type MMTV integration site family, member 1 (WNT1); INT1; Official Gene Symbol: WNT1 Gene ID: 7471 (Human) Gene Map Locus: 12q13 (Human) Wnt1, a 40KDa cyteine-rich secretory glycoprotein, is a highly conserved archetypal member of WNT protein family. Wnt1, originally identified as proto-oncogene, plays a significant role in oncogenesis and in various developmental processes including regulation of cell fate, cell survival and patterning during embryogenesis. Wnt1 binds to its FZD receptor on the cell surface resulting in activation of beta-Catenin and translocation into the nucleus. Activated beta-catenin makes a complex with TCF family of transcription factors in the nucleus resulting in transactivation of several genes including c-myc and Cyclin D1 who play pivotal role in cell survival and differentiation. Mutations in the members of Wnt1 signaling have been suggested in several cancer types.
